differentiation of iranian strains of brucella spp. by random amplification of polymorphic dna
Authors
abstract
101 isolates of brucella spp. including the reference (n=7) and the iranian field strains belonging to last years 1961-2003 (n=94) were biotyped and classified into 9 biotypes. random amplified polymorphic dna (rapd) analysis using a ten mer-ap4 primer was generated 72 reproduciable dna band. the typeability was 100%. the thirteen isolates were classified into five groups, each containing 2-4 biotypes. the high dna polymorphisms among the strains suggested that animals in iran have been infected with genetically diverse isolates however, in the last forty years b.abortus biotype a3 and b.melitensis biotype m1 have been predominantly isolated from brucellosis cases. the results indicated that the rapd analysis using ap4 primer can be used to attain reproducibility amplify random fragments of dna from brucella genomes in order to differentiate at species and sub-species for epidemiological purpose.

full textDifferentiation of Aspergillus ustus strains by random amplification of polymorphic DNA.
The sodium dodecyl sulfate-polyacrylamide gel electrophoresis patterns of water-soluble proteins and the randomly amplified polymorphic DNA (RAPD) patterns of whole-cell lysates from 21 Aspergillus ustus isolates, including 11 reference strains and 10 patient and environmental strains from one hospital, were investigated. All isolates showed identical protein patterns. The RAPD assay discrimina...
full textStrain differentiation of Trichophyton rubrum by random amplification of polymorphic DNA (RAPD).
Trichophyton rubrum is an important cause of dermatomycoses. Molecular strain typing methods have recently been developed to address questions about epidemiology and source of relapse following treatment. This report describes the application of RAPD for molecular strain differentiation of this fungus utilizing the primers 1- (5'-d[GGTGCGGGAA]-3') and 6- (5'-d[CCCGTCAGCA]-3'). A total of five R...
full textDifferentiation of Aspergillus parasiticus from Aspergillus sojae by random amplification of polymorphic DNA.
Aspergillus parasiticus and Aspergillus sojae are two morphologically similar species belonging to the Aspergillus section flavi. A new method to distinguish the two species, the aflatoxin producer A. parasiticus and the koji mold A. sojae, was developed. Single primers with arbitrary sequences were used to generate random amplified polymorphic DNA (RAPD) markers from strains of these two speci...
